HE550AZ medium is a chemically defined, fully synthetic (serum-free) medium designed with a new concept for suspension culture (expression and production) of human embryonic kidney (HEK) 293 cells, such as Expi293F Pro, Expi293F, 293-F, HEK293, 293T, and VPC cells.
HE550AZ medium represents our flagship formulation, created to enable the next stage of high-density production culture of 293 cells.
HE550AZ medium contains no protein, hydrolystates, animal-derived components, human-derived components, macromolecules, or phenol red, ensuring improved lot-to-lot consistency. These features facilitate downstream purification processes for recombinant proteins and contribute to robust and reproductive virus production process.
Because HE550AZ medium dose not interfere with gene delivery or gene expression, it is designed to support high-efficiency transfection and scalable production of recombinant proteins and viral vectors.
HE550/HE550AZ medium was used for shaker culture in disposable Erlenmeyer flasks under the following conditions.
HE550/HE550AZ medium supports efficient cell proliferation of Expi293F Pro cells, comparable to Expi293 Pro Expression medium from Thermo Fisher Scientific.
*Expi293F Pro cells that had been cultured in Expi293 Pro Expression medium were adapted to the HE550AZ medium prior to use.
＜ Culture conditions ＞
(1) Seeding cell density：3 x 10e5 cells/mL.
(2) Shaker culture：125 rpm, 37℃, 8% CO2.
(3) Culture volume：30 mL with L-alanyl-L-glutamine.
HE550/HE550AZ medium was used for shaker culture in disposable Erlenmeyer flasks under the following conditions.
HE550/HE550AZ medium provides the best cell growth compared with our CD media.
＜ Culture conditions ＞
(1) Seeding cell density：3 x 10e5 cells/mL.
(2) Shaker culture：125 rpm, 37℃, 8% CO2.
(3) Culture volume：30 mL with L-alanyl-L-glutamine.
HE550/HE550AZ medium was used for shaker culture in disposable Erlenmeyer flasks under the following conditions.
HE550/HE550AZ medium provides the best cell growth compared with our CD media.
*HEK293 cells cultured in serum-containing medium were adapted to HE400AZ medium for suspension culture prior to use.

(2) Shaker culture：125 rpm, 37℃, 8% CO2.
(3) Culture volume：30 mL with L-alanyl-L-glutamine.
